The immune system has 2 Methods of Protecting folks from microorganisms including bacteria, fungus, protozoa, mycoplasmas and viruses.
1. The Cell mediated Immunity where white blood cells also called lymphocytes fight all types of microbes. These are many types- macrophages, microglia, T cells, plasma cells, etc.
2. Humoral Immunity - involving special substances or chemicals the body produces called the humors or fluid part of the blood - some of these chemokines include Interferon, Interleukins, various antibodies,
tumor necrosis factors, complement and more.
In other diseases such as Rheumatoid arthritis, both the Cell Immunity and humor immunity are actively involved in autoimmune processes, but compared to AS, lymphocytes do not attack the spine.
Research found activity by complement; Complement are proteins – our immune system produces them and they are good at attacking and destroying invading microorganisms including viruses. Normally complement are safe and do not cause problems or damage to parts of the body.. But in Ankylosing spondylitis, the complement is implicated to start the attack.
The following are observed in Ankylosing spondylitis:
1. Immunoglobulins – also known as antibodies> Immunoglobins G-IgG, Immunoglobin A - IgA and Immunoglobin M -IgM
2. Complement protein ( immune reactive factors )- have names beginning with C for example C1, C3, C7
Complement – there are several of them but together they are effective at killing and destroying bacteria and other microorganisms. If one were to observe complement at work , they usually attach to the cell walls, for example bacteria and form tiny holes or breaks on the walls. But complement is unique because when it acts, there are several of these holes and they do not close nor repair. When that happens to bacteria, the holes or breaks leads to leaking of the contents of the cell and that cell is doomed and eventually dies.

One method suggested by medical research is to treat AS using Complement Inhibitors.
But that therapy will not work for 2 reasons.
First, the liver produces complement and these circulate in the blood and will flow into body tissues including the spine. Macrophages and lymphocytes also produce complement - they become attracted to parts of the body where there is even a slight sign
of inflammation and is followed by more and more lymphocytes to the same area to start an attack.
Second, an experiment with complement inhibitor Soliris ( eculizumab ) resulted in negative side effects - infections became common. This will not work.. There is a need to find other methods to remedy AS.
Complement can be present in the blood and even in high levels but should not pose problems to the host since it normally appears in an inactive form.
What activates Complement?
There are 3 pathways
1. Classical pathway
2. alternative pathway
3. mannose binding pathway.
For the explanation, we should only consider Classical pathway because it is the one that is involved in the case of Ankylosing spondylitis. It is Antibodies – specially Immunoglobulin G and Immunoglobulin M. When antibodies attaches or binds to an antigen, it becomes an Antigen-Antibody complex & changes and is the kind that will activate complement if complement is present.

The articles above shows all 3 are present – 1. the target Antigens Collagen II & Collagen IV –2. antibody and 3. complement. The complement starts destruction of 2 collagens found in cartilage and that is how AS disease starts and progresses.
The Immune System has its own controls:
1. Helper T cells functions to increase antibody production.
2. Suppressor T cell or Regulatory T Cells - gives the signal to lower antibody production.
If a method were found that is able to harness the T Reg cells, it will be the solution to so many autoimmune diseases.

But Oral Tolerance is not successful in humans.
The way to Success is a new discovery which is B7.
The Research by Dr. Polly Matzinger explains the T cells requirements.
The T cell needs 2 factors or signals for full activation.
1.) a peptide of the 2 target antigen - Collagens II & IV, – the dose primes or makes ready the T cell. But without the 2nd signal the T cell remains in anergy ( no activity).. If both collagens are accepted by the T Reg cell, it will lower antibodies specific to collagens ONLY. It is very specific.
2.) the Second signal is co-stimulation –which is B7 also known as CD80 (B7-1) & CD86(B7-2) – either one is enough to make the T cell go through the changes that makes it a fully activated cell.

Dr. Matzinger’s discovery is called Danger Theory because methods available to Modern Medicine to induce B7 were Vaccines, Toxoids & byproducts from dangerous micro-organism & not applicable due to toxicity including inflammatory lymphokines, high fevers, fatigue from an overactive immune system ..
But Dr. Matzinger was not aware there is a nontoxic and safe method to induce B7 and that is through Herbs.
However, Modern Medicine has the policy of not including herbs with their drugs. They feel that herbs are cheap and cheap means little or no profit.

How the formula works.
Ankylosing Spondylitis -there are high antibodies in patients caused by helper T cells.
Oral Tolerance leads to proliferation of new Suppressor T cells, the new cells work against Helper T cells which delete each other, when Helper T cells are reduced, high antibodies to Cartilage collagens go down to normal, each course leads to improvement until complete healing.
