What causes Alzheimer's Disease (AD)?

New information shows there is inflammation &  active immune processes in Alzheimer's patients brains.

Together with amyloid plaques, the presence of the following are observed in Alzheimer's disease:

The immune system protects the body via
1.  lymphocytes – white blood cells of many types- macrophages, microglia, T cells, plasma cells, etc.
2. Humoral Immunity - involving chemokines in the humors or fluid part of the blood.

In other diseases such as Rheumatoid arthritis, both the lymphocytes and humor immunity are involved, but comparing this to AD, lymphocytes do not attack brain or cells.

Research found activity by complement; Complement are proteins – our immune system produces them and they are good at attacking and destroying invading microorganisms including viruses. Normally complement are safe and do not cause problems or damage to the brain. But in Alzheimer's disease the complement is implicated to start the attack.


The following are observed in Alzheimer's disease:

        1.  Immunoglobulins – also known as antibodies
        2.  Complement protein ( immune reactive factors )- have names beginning      with C for example C1,  C3, C7

        3.  microglia and macrophages –   blood cells which are normal parts of             the immune system . Microglia are known as resident lymphocytes , these                     cells stay within the regions of the brain & act as protection against all                           pathogens, germs, bacteria, fungi & viruses.
         4. Astrocytes –  part of the brain cells.

 



Complement – there are several of them but together they are effective at killing and destroying bacteria and other microorganisms.   If one were to observe complement at work , they usually attach to the cell walls of for example bacteria and form tiny holes or breaks on the walls.

But complement is unique because when it acts, there are several of these holes and they do not close nor repair. When that happens to bacteria, the holes or breaks leads to leaking of the contents of the cell and that cell is doomed and eventually dies.



The complement can be found at the earliest stages of amyloid deposition and their activation coincides with the clinical expression of Alzheimer's dementia.  There is increased production & levels of complement in the blood.

One strategy suggested by medical research is to treat AD using Complement Inhibitors. 
 
But that therapy will not work for 2 reasons.


First, the macrophages, microglia and even astrocytes which are on areas that the Alzheimer's condition starts also produce complement.

Second, the Alzheimer brain apparently mounts a defense against chronic complement attack.

The following are Complement Inhibitor factors:

  1. clusterin 

  2. C1-inhibitor 

  3. lactoferrin 

  4. C4 binding protein 

     

    All 4 above are upregulated in AD. As mentioned, the brain does produce these Complement inhibitors but it does not fix the problem

    Higher levels of these Complement inhibitors are found in the AD brain, C1-inhibitor is detected in its cleaved form in AD.  

    Presence of the cleaved form, therefore, indicates that C1 has been activated and then inhibited by the C1-inhibitor

     

     Since these do not prevent the disease, there is a need to find other methods to remedy AD..





What exactly is being attacked by the immune system?

It is the vimentin in neurons.


The 2 articles below points to the protein vimentin which is the target antigen in Alzheimer's Disease.

Other journals also states vimentin is attacked in AD but 2  should be sufficient to prove
our point.

Complement can be present in the blood and even in high levels but should not pose problems to the host since it normally appears in an inactive form.


What activates Complement?

 

There are 3 pathways

  1. Classical pathway

  2. alternative pathway

  3. mannose binding pathway.


 

For the explanation, we should only consider Classical pathway because it is the one that is involved in the case of Alzheimer's disease.  It is Antibodies – specially Immunoglobulin G and Immunoglobulin M. When antibodies attaches or binds to an antigen, it  becomes an Antigen-Antibody complex  & changes and is the kind that will activate complement if complement is present.


Our article here explains all 3 are present – vimentin – antibody and complement. The complement starts destruction of brain cells and that is how alzheimers disease starts and progresses.

How do these all relate to the amyloid?
 GFAP and Vimentin surround a core which is amyloid in astrocyte cells.  The immune attack is on vimentin  which releases its covering and the amyloid.  The amyloid is not attacked and accumulates as debris.





‭The Immune System has its own controls:


‭1.  Helper T cells functions to increase antibody production.


‭2. Suppressor T cell or Regulatory T Cells - gives the signal to lower antibody production.





If a method were found that is able to harness the T Reg cells, it  will be the solution to so many autoimmune diseases.

How to harness the T Reg Cell


The way to use T Reg cell‭ ‬-‭ ‬the method is Immune Tolerance and Oral Tolerance.

Immune Tolerance means tolerating Antigens instead of attacking them.

Oral tolerance is the suppression of immune responses to antigens‭ (‬Ag‭) ‬that have been administered previously by the oral route.‭ ‬Multiple mechanisms of tolerance are induced by oral ‭ ‬Antigen.‭ ‬Low doses favor active suppression,‭ ‬whereas higher doses favor clonal anergy/deletion.

But Oral Tolerance is not successful in humans.


The way to Success is a new discovery which is B7.

The Research by Dr.‭ ‬Polly Matzinger explains the T cells requirements.

The T cell needs‭ ‬2‭ ‬factors or signals for full activation.

1.‭) ‬a peptide of the target antigen‭ ‬-‭ ‬vimentin,‭ – ‬the dose primes or makes ready the T cell.‭ ‬But without the‭ ‬2nd signal the T cell remains in anergy‭ ( ‬no activity‭)‬..  If vimentin is accepted by the T Reg cell, it will lower antibodies specific to vimentin ONLY.  It is very specific.

2.‭) ‬the Second signal is co-stimulation‭ –which ‬is B7‭ ‬also known as CD80‭ (‬B7-1‭) & ‬CD86‭(‬B7-2‭) – ‬either one is enough to make the T cell go through the changes that makes it a fully activated cell.

Dr.‭ ‬Matzinger’s discovery is called Danger Theory because methods available to Modern Medicine to induce B7‭ ‬were Vaccines,‭ ‬Toxoids‭ & ‬byproducts from dangerous micro-organism‭ & ‬not applicable due to toxicity including inflammatory lymphokines,‭ ‬fevers,‭ ‬fatigue from an overactive immune system‭ ‬..

But Dr.‭ ‬Matzinger was not aware there is a nontoxic and safe method to induce B7‭ ‬and that is through Herbs.


However, Modern Medicine has the policy of not including herbs with their drugs.  They feel that herbs are cheap and cheap  means little or no profit.

DIRECTIONS FOR TAKING FORMULA:
